synthesis method | 1, 1-dimethylethyl (2S)-2-methyl-1-piperazine carboxylate (step a) solution and n-BuLi(25 mL) stored in THF(200 mL) at room temperature, 1.6 M stored in hexane, 40 mmol) mixed. Stir the solution for 30 min, then add TBDMSCl(3.04 g,20mm ol). Stir the mixture for an extra hour and add (Boc)2O(5.2g,24 mmol) to the solution. The resulting mixture was stirred for one hour and diluted with H2O(50 mL). The organic layer was separated, washed with salt water (50 mL), dried on Na2SO4 and concentrated under vacuum. Rapid chromatography on silica gel (5% MeOH/2% NH4OH/93% CH2Cl2) provided yellow oily title compounds (3.7 g,93%). (S)-1-N-Boc-2-methylpiperazine, yield (3.7g,93%).
